本研究利用本所分離的虹彩病毒及神經壞死病毒病毒株,分別選取90-點帶石斑株之虹彩病毒及94-471 棕點石斑株之神經壞死病毒為疫苗株,進行宿主感受性測試並測定其半致死劑量(LD50),以決定其攻毒劑量,評估其不同之不活化條件、搭配不同佐劑、不同的抗原與佐劑比及不同之免疫途徑,以研發單價不活化病毒疫苗,評估疫苗之效力。結果顯示,在宿主感受性試驗發現金目鱸、紅魚及點帶石斑對虹彩病毒都非常敏感,其LD50力價分別位於104.5 TCID50/mL、106.5 TCID50/mL及105. 9 TCID50/mL。神經壞死病毒之敏感性宿主試驗發現金目鱸及點帶石斑其LD50力價分別為104.6 TCID50/mL及106.9 TCID50 /mL。在不活化病毒試劑比較結果,以0.1%福馬林於4℃不活化虹彩病毒之疫苗效力之保護指數為60%較37℃不活化之疫苗效力為高。在不同佐劑及投予方式之比較結果發現,單價點帶石斑虹彩病毒不活化疫苗其混以佐劑B 之疫苗效力之保護指數為53.4%較佐劑A 及佐劑C 為高;以腹腔注射混有佐劑B 之疫苗,其保護指數為73%;混有MVP佐劑疫苗之保護指數只有50%;腹腔注射的免疫效果較肌肉注射效果為佳。在單純溫度不活化結果顯示,單價棕點石斑神經壞死病毒不活化疫苗之初步結果發現,以37℃不活化之疫苗效力較56℃及70℃不活化為佳。

In this project, we select the field virus with high virulence and antigenecity as the seed virus for vaccine, strain 90-grouper for iridovirus and strain 94-471 for nervous necrosis virus. To evaluate the efficacy of vaccine, we determine the half lethal dose of suspected hosts like sea bass, grouper, crimson snapper to decide challenge dose, different combination of adjuvants and routes of administration for development an univalent or bivalent vaccine. The results shows the suspected hosts like sea bass, grouper and crimson snapper all sensitive to iridovirus, whose LD50 for sea bass, grouper and crimson snapper were to 104.5TCID50/ml, 106.5TCID50/ml and 105.9TCID50/ml respectively. In nervous necrosis virus, the suspected hosts were sea bass and grouper, whose LD50 were 104.6TCID50/ml and 106.9 TCID50 /ml respectively. We found 0.1% formalin inactive vaccine at 4℃had 60% protective index higher than at 37℃; the efficacy of univalent inactive iridovirus vaccine with adjuvants B is better than with adjuvants A and C. The protective indexes in vaccine mixed with adjuvants B and delivered by intraperitoneal injection is 73%; the vaccine mixed with MVP adjuvants and delivered by intramuscularly injection is 50%. The route of vaccine delivery by intraperitoneal injection is better than intramuscularly injection. The preliminary results for the efficacy of univalent inactive nervous necrosis virus or bivalent vaccine indicated that the inactivation of virus at 37℃was better than at 56℃and 70℃.

我國石斑魚苗養殖業目前成長受限於石斑虹彩病毒感染症(Grouper Iridovirus Infection)及神經壞死病毒(Nervous necrosis virus, NNV),損失高達八成〔1,2〕。此兩種病毒皆會造成二十餘種之魚類感染且使現今養殖產業變成疾病發生高風險之產業。為提昇養殖魚類之產量又兼顧優良、安全之生產品質,以發展疫苗控制上述疾病成為唯一的對策。日本針對嘉魶魚之虹彩病毒已有不活化單價疫苗及雙價疫苗上市,作為防疫之策略。因此,本研究擬利用本所自行分離的虹彩病毒及病毒神經壞死病毒株,測試病毒不活化條件及搭配不同佐劑研發單價或二價不活化病毒疫苗,評估疫苗之效力以及免疫的最佳投予時間,期待能研發出具有經濟效益、穩定有效益的疫苗,也能立即解決現今養殖業的窘境,並提昇養殖產業之國際競爭力。
